Energy: Platinum Power Resumes Normal Activity

Immune-stimulatory Activity with RGX-104 in Advanced Cancer Patients

Rgenix, Inc., a clinical stage biopharmaceutical company developing first-in-class small molecule and antibody cancer therapeutics, announced today preliminary data from an ongoing Phase 1a/b clinical trial with its lead oral investigational agent, RGX-104. These data demonstrate immune-stimulatory activity in solid tumor patients with highly-refractory malignancies, including patients who have failed prior checkpoint inhibitors. Also presented were pre-clinical data establishing the immune-modulatory and anti-tumor effects of RGX-104. The company presented the data at the AACR-NCI-EORTC International Conference on Molecular Targets and Cancer Therapeutics in Philadelphia. RGX-104 is a liver X receptor (LXR) agonist that upregulates the expression of the target gene, Apolipoprotein E (ApoE), triggering several downstream effects via ApoE receptors. In pre-clinical data presented today, treatment with RGX-104 in mouse models resulted in dual effects on myeloid-derived suppressor cells (MDSCs) and dendritic cells (DCs), both innate immune cells that play a central role in regulating anti-tumor immunity and response to checkpoint inhibitors. Innate immune activation with RGX-104, coupled with a reduction in tumor blood vessels, resulted in anti-tumor activity as a monotherapy as well as synergy with checkpoint inhibitors (CPI) in several drug-resistant mouse models. These data provide rationale for Rgenix’s ongoing Phase 1a/b trial of RGX-104 in advanced cancer patients and support evaluation of RGX-104 as both a monotherapy as well as in combination with CPIs.
As part of the ongoing Phase 1a/b clinical trial, 15 patients with a variety of solid tumors have been treated with escalating doses of RGX-104 monotherapy. Patients treated with RGX-104 had a median of six prior therapies with a range of 1-12, highlighting a population of patients with profoundly resistant disease.Activation of the LXR-ApoE pathway with oral administration of RGX-104 was associated with immune-stimulatory activity in 9 of 10 evaluable patients. This was demonstrated by an increase (up to 11-fold) in activated circulating PD-1+CD8+ T cells during treatment. T cell activation was observed in patients who experienced modulation of the innate immune system during treatment. The effect of RGX-104 on the innate immune system consisted of both MDSC depletion (up to 95% decrease) as well as DC activation as indicated by induction of PD-L1 expression (up to 100% increase). In most cases these effects were observed within two weeks of treatment initiation and generally preceded the onset of T cell activation.
Safety data demonstrate good tolerability with on-target safety findings in the first three dosing cohorts. One patient experienced a DLT of grade 4 reversible neutropenia – a known potential effect of LXR agonism – that reversed within one week, allowing the patient to subsequently tolerate a 50% dose reduction. No MTD has been reached to date. Stable disease has been observed in 4 of 12 evaluable patients, including three who have failed prior checkpoint inhibitor therapy, for periods of at least 8 weeks.Rgenix plans to enroll subsequent dose-escalation cohorts of the RGX-104 monotherapy trial in Q4 2017. Additionally, Rgenix is planning to initiate the Phase 1b expansion component of the study, comprised of disease directed cohorts receiving RGX-104 monotherapy as well as cohorts receiving RGX-104 combined with a CPI, projected to begin in 1H 2018.The LXR-ApoE pathway was discovered as a cancer target using a microRNA (miRNA) based target discovery approach originally developed at The Rockefeller University and now exclusively licensed to Rgenix.

Nel sistema nervoso centrale, i neuroni formano una rete stereotipata di connessioni sinaptiche altamente specializzate che permettono il trasferimento di informazioni con alta precisione spaziale e temporale. Tale rete di connessioni svolge un ruolo fondamentale nel processo e nell’integrazione delle informazioni sensoriali provenienti dal mondo esterno.
Un numero crescente di evidenze sta mostrando come cambiamenti del funzionamento cerebrale – sia durante condizioni fisiologiche che patologiche – siano associati a cambiamenti dinamici dei circuiti neuronali. Durante le fasi dello sviluppo, i circuiti neuronali vanno incontro a uno stadio caratterizzato dall’eliminazione di connessioni sinaptiche aberranti rafforzando invece circuiti critici per il funzionamento cerebrale. Inoltre, i fenomeni di plasticità sinaptica determinano un cambiamento della connettività e del funzionamento dei circuiti cerebrali che garantiscono il corretto funzionamento delle funzioni superiori del cervello, come la memoria e l’apprendimento, per tutta la durata della vita. D’altro canto, alterazioni dei circuiti sinaptici possono verificarsi anche durante stati patologici cerebrali portando a degenerazione del cervello.
Lo sviluppo di recenti tecnologie che consentono di manipolare geneticamente e funzionalmente specifici sottopopolazioni di neuroni (e.g. targeting genico, optogenetica e farmacogenetica) stanno fornendo un’opportunità senza precedenti per studiare il ruolo di questi neuroni e delle connessioni sinaptiche in precisi circuiti cerebrali. 20 years of Europe! Our activity report 1996-2016

europe“It is a great honour for me to introduce this Report looking back over the twenty years of activity of the European think tank founded by Jacques Delors in 1996, especially as I have just become its President. This Report presents the main European issues and challenges on which the Institute has focused steadfastly since its foundation, through calmer periods and through crises, in order to promote European construction in accordance with Jacques Delors’ legacy and vision. It provides an impressive overview of the actions undertaken with European decision-makers and the general public, which acts as an important stimulus to pursue our work over the coming months and years. It goes without saying that the current questioning of the EU is an additional motivation to contribute to its revival with even more determination. The first three sections of this Report demonstrate how the Institute has worked tirelessly to “Reconcile the EU and its citizens”, “Combine competition, cooperation and solidarity” and “Bolster the EU’s role in the globalisation process”. A few examples of our publications, events and media appearances are given, all of which aim in particular to consolidate the European Federation of Nation States, keep European democracy alive, promote the right to freedom of movement and the Schengen area, deepen the Single Market, complete the Economic and Monetary Union, step up Social Europe, create a genuine “Energy Union”, think global and act European, improve relations between the EU and its neighbours, etc. I would, of course, recommend that you look into these many analyses and recommendations more closely by visiting our website The fourth section of this Report sets out the remarkable results achieved in terms of publications, the conferences and seminars we host or attend, our presence in European and international media and the number of our fans and followers. 3 out of 4 startups are acquired by US companies

startupsSan Francisco. US companies are more acquisitive. Even less surprisingly, among the top 15 acquirers, over 70% are Silicon Valley companies. This is the main finding from “Startup Transatlantic M&As. US vs EU”, the report that will be presented today at the European Innovation Day conference in Mountain View, the event created and organized by Mind the Bridge and co-organized by EIT Digital.The research completed by Mind the Bridge and CrunchBase analyses approximately six thousand startup acquisitions performed by US and European companies since 2012. It reveals that US and European companies focus their M&A activity in the US and Europe: 91% of the acquisitions performed by US and European companies – and 96% of the price paid – refer to US and European startups. “The common wisdom is that acquisitions have played a central role in Silicon Valley’s success, and that buying startups is one of the fastest ways for companies to embrace disruption and keep innovating – Alberto Onetti, Chairman Mind the Bridge, comments – The aim of this report was to understand if these beliefs are actually true. Now the numbers speak for themselves.”In particular, US companies acquired a number of startups that is significantly higher than EU companies, approximately 4 times more. 4,880 deals out of 5925, in fact, have been performed by US companies representing 82% of all the transatlantic transactions and investing $504 billion in disclosed transactions. This represents 90% of the overall value invested in startup acquisitions across the Atlantic. Silicon Valley alone registers 1,264 deals (21% of all the transatlantic deals and 26% of US ones) for $118B, more than the whole Europe. The Old Continent accounts for 1,045 deals – 18% of the total – for $57B. The median deal size of US acquisitions is $110M, 1.83 times than the median amount invested by EU companies ($60M). Additionally, if we look at the distinction between domestic and cross-atlantic acquisitions of startups, it has been reported that US buys prevalently domestic companies (4,300 out of 4,900 total EU/US based startups acquired, 88% on the number of acquisitions, 80% of the value of disclosed transactions). 68% of European startups acquisitions (700 out of the 1,045 total) happen domestically.
To be noted that US startups accounted for just under a third of the number of European startup acquisitions but ate up 59% of the total capital invested. This data suggests, on the one hand, that there is still a gap to be filled in the European startup ecosystem’s ability to produce exits; on the other hand – even more interestingly – US startups are more expensive than their European counterparts (32% of the acquisitions absorbed 59% of the overall amount paid).
“This is the first report doing a comparative analysis of acquisitions looking at Europe and the US. The interdependence of Europe and the US is striking and second only to the dependence of European countries on each other. The findings will inform corporate strategy in both continents”, says Gené Teare Head of Content at CrunchBase.
startups1Remarkably, US companies acquired 562 European startups while 709 startups were acquired by European companies in Europe. That means – consistently with data from SEP Monitor – that 44% of European startup acquisitions have been performed by US companies.
Research introduces the “exits/acquisition ratio” that shows the ratio between the number of exits and the number of acquisitions. Not all ecosystems have a positive balance between the sell (exits) and buy (acquisitions) side. US shows a certain equilibrium with 0.95 while Europe is characterized by a negative M&A balance (1.2): it means that the overall number of startups that have been sold in a region is larger than the ones that have been acquired by regional companies. This situation is clearly evident in the youngest ecosystems (Spain, Italy, the Benelux).
When broken down by city, New York, Chicago and some municipalities of the Silicon Valley (including Mountain View, Sunnyvale and San Jose), and to a lesser extent Los Angeles, Austin and Boston are “net acquirers” of startups (the number of startups acquired is larger than the ones that have been sold), while San Francisco, Seattle and Palo Alto are “net sellers” of startups. Silicon Valley, when considered as a whole, demonstrates a healthy balance (0.95), slightly below the parity.
US and Silicon Valley Run the M&A Show – Not surprisingly the top 15 acquirers in the transatlantic ranking are all US companies. Even less surprisingly, 11 out of the top 15 are from the Silicon Valley. The first European one – Germany’s SAP – ranks 33rd. On average, the top 15 US-based companies acquire 6 times more startups than the top European companies. Moderate physical activity linked with 50% reduction in cardiovascular death in over-65s

cardiology congress-2016Rome, Italy – 27 Aug 2016 Fiera di Roma (Ingresso Nord, Via Portuense 1645/1647): Moderate physical activity is associated with a greater than 50% reduction in cardiovascular death in over-65s, according to research presented at ESC Congress 2016 today.1 The 12 year study in nearly 2500 adults aged 65 to 74 years found that moderate physical activity reduced the risk of an acute cardiovascular event by more than 30%. High levels of physical activity led to greater risk reductions.“The role of physical activity in preventing cardiovascular disease (CVD) in people of working age is well established,” said Professor Riitta Antikainen, professor of geriatrics at the University of Oulu, Finland. “But relatively little is known about the effect of regular physical activity on CVD risk in older people.”The present study assessed the association between leisure time physical activity and CVD risk and mortality in 2456 men and women aged 65 to 74 years who were enrolled into the National FINRISK Study between 1997 and 2007.Baseline data collection included self-administered questionnaires on physical activity and other health related behaviour, clinical measurements (blood pressure, weight and height), and laboratory measurements including serum nuova-fiera-di-romacholesterol. Participants were followed up until the end of 2013. Deaths were recorded from the National Causes of Death Register and incident CVD events (coronary heart disease and stroke) were collected from the National Hospital Discharge register.The researchers classified self-reported physical activity as:
• Low: reading, watching TV or working in the household without much physical activity.
• Moderate: walking, cycling or practising other forms of light exercise (fishing, gardening, hunting) at least four hours per week.
• High: recreational sports (for example running, jogging, skiing, gymnastics, swimming, ball games or heavy gardening) or intense training or sports competitions at least three hours a week.
During a median follow-up of 11.8 years, 197 participants died from CVD and 416 had a first CVD event.

Fraunhofer researchers on SmartHome, Lifestyle and Industry 4.0: “Potential of activity recognition is far from being exhausted”

Acceleration sensors in smartphones, smartwatches and in clothes are making the unobtrusive recognition of physical activity possible. In sports, they help digital trainers correct exercises. Intelligent living environments recognize falls and call for help. A hand movement can dim the light, lower venetian blinds or turn on the TV.Such technologies are developed at Fraunhofer IGD. Technologies recognizing a person’s activity, thus allowing us to control applications and machines. For instance, they will measure your pulse, body movements or blood pressure. These data help physicians and therapists better evaluate the state of health and future development of a patient. At the workplace, an unhealthy body posture, excessive physical strain or dangerous noise levels can be detected. In the process, the Fraunhofer researchers attach great importance to the security of the collected data.According to Professor Bodo Urban of Fraunhofer IGD in Rostock, we can expect exciting developments in the industry in the years to come: “Activity recognition is taking man-machine interaction to the next level.” Machines and robots recognize human activities and respond to these. Production plans can be intuitively controlled by gestures via a smartwatch. For instance, lifting up your hands in the case of a malfunction will trigger the emergency switch. As energy prices across the world remain volatile, it’s become more important to explore new sources of oil and gas to help alleviate the stress on consumers and businesses. Unless, of course, you’re Greenpeace. Fresh from their successful campaign to halt the Keystone XL Pipeline in the U.S., Greenpeace is now taking action in the United Kingdom to encourage government ministers to drop their support for companies such as Cairn Energy who would like to pursue more oil drilling in the Arctic. Speaking before a parliamentary committee regarding Cairn Energy’s activities is clearly a major change for Greenpeace, given that they have previously resorted to criminal activities in Greenland.Just last year four Greenpeace activists “climbed aboard Cairn Energy’s Stena Don platform off the island’s coast and rigged up hanging tents.” The government of Greenland criticized Greenpeace for its “very grave and illegal attack,” before highlighting its sovereign right to explore “opportunities to secure the economic foundation for its people’s condition of life.” As we have documented in Indonesia and Papua New Guinea, Greenpeace has no respect for state sovereignty. The statement from Greenland’s government could be any country describing their ongoing travails against Greenpeace. British parliamentarians providing yet more publicity to an organization that thrives on criminal activity is an insult to those across the world who have fought Greenpeace for their rights to pursue economic opportunity.

Broward County Reports Strong Sales Activity in September

Miami, FL – In Broward County, single-family home sales increased 11 percent, from 974 in September 2010 to 1,079 last month, according to the Broward Council of the 25,000-member MIAMI Association of REALTORS and the local Multiple Listing Service (MLS) systems. Condominium sales increased six percent from 1,214 to 1,281.
Statewide sales increased 10 percent to 15,036 for single-family homes and 10 percent to 6,666 for condominiums compared to September 2010. Nationally, sales of existing single-family homes, townhomes, condominiums, and co-ops declined three percent from the previous month and but were 11.3 percent above September 2010, according to the National Association of Realtors (NAR).“The Broward County real estate market continues to perform in a healthy and balanced manner, as local home prices stabilize or rise in many areas and neighborhoods,” said Terri Bersach, 2011 president of the Broward County Board of Governors of the MIAMI Association of REALTORS. “Inventory is declining, as housing demand exceeds available supply. International buyers, who mostly pay all cash, are playing a major role in market strengthening as evidenced by the high number of cash condominium sales.”

The NSC has been in operation since 2001 and is the Alliance’s primary point of contact for commercial shipping. At present, its main focus in NATO’s counter piracy mission, Operation Ocean Shield. Since its inception, the NSC has participated in several NATO maritime operations such as Operation Active Endeavour (counter terrorism in the Mediterranean) and Operation Unified Protector off the coast of Libya. Nato continues to disrupt pirate activity

It was yet another busy night for the Danish warship Hdms Abaslon in the Somali Basin the night between the 1st and 2nd of March 2010 where the ship again located, intercepted and disposed of two more “whalers” the so called mother skiffs used by pirates as logistic platforms to transport their attack teams offshore. At the request of the UN in late 2008, NATO started to provide escorts to UN World Food Programme vessels transiting through the Somali Basin. This operation has now been succeeded by Operation Ocean Shield, which as well as deterring and disrupting pirate activity additionally offers training to regional countries in developing their own capacity to combat piracy activities.  NATO is conducting counter-piracy activities in full complementarity with the relevant UN Security Council Resolutions (1816, 1838, 1846, 1851 and 1897) and in coordination with other international actors including the European Union.
