Late-Breaker Session at EADV Congress

Posted by fidest press agency su martedì, 15 ottobre 2019

Concert Pharmaceuticals, Inc. (NASDAQ: CNCE) today presented results from its recently completed Phase 2 dose-ranging trial of the investigational medicine CTP-543 in patients with moderate-to-severe alopecia areata, an autoimmune disorder that results in patchy or complete hair loss. Patients in the study treated with either 8 mg twice-daily or 12 mg twice-daily of CTP-543 met the primary efficacy endpoint with statistically significant differences (p <0.001) relative to placebo in the percentage of patients achieving a ≥ 50% relative change from baseline at 24 weeks using the Severity of Alopecia Tool (SALT). These results were presented during the 28th European Academy of Dermatology and Venereology (EADV) Annual Congress in Madrid, Spain. In the Phase 2 trial, the primary efficacy endpoint in the 12 mg twice-daily cohort was met with 58% of patients achieving a ≥ 50% relative reduction in their overall SALT score from baseline compared to 9% for placebo (p <0.001), with statistically significant separation from placebo occurring at Week 12. In the 8 mg twice-daily cohort, 47% of patients achieved the primary endpoint compared to placebo (p <0.001). For the 4 mg cohort, 21% of patients achieved a ≥ 50% relative reduction in their overall SALT score from baseline, however there was not a statistically significant difference from placebo.Data from the Patient Global Impression of Improvement scale showed 78% of the patients receiving 12 mg twice-daily and 58% of the patients receiving 8 mg twice-daily rated their alopecia areata as “much improved” or “very much improved” at Week 24. For both doses there was a statistically significant difference from placebo (p <0.001).In the Phase 2 trial, the most common (≥10%) side effects in the 12 mg CTP-543 dose group were headache, nasopharyngitis, upper respiratory tract infection and acne. One serious adverse event of facial cellulitis was reported as possibly related to treatment. After a brief dosing interruption, treatment was continued and this patient completed the trial. No thromboembolic events were reported during the trial. The Phase 2 trial was a double-blind, randomized, placebo-controlled, sequential dose trial to evaluate the safety and efficacy of CTP-543 in adult patients with moderate-to-severe alopecia areata. A total of 149 patients were randomized to receive one of three doses of CTP-543 (4 mg, 8 mg, or 12 mg) or placebo, administered twice-daily. The primary outcome measure utilized the SALT score after 24 weeks of dosing. The average baseline SALT score across all patients was approximately 88%, where 0% is no scalp hair loss and 100% represents total scalp hair loss. All patients who completed 24 weeks of treatment in the 12 mg dosing cohort had the opportunity to continue in a separate extension study to evaluate long-term safety and efficacy of CTP-543.Details from the oral presentation, entitled “CTP-543, an oral JAK inhibitor, achieves primary endpoint in Phase 2 randomized, placebo-controlled, dose ranging trial in patients with moderate-to-severe alopecia areata,” is available in the Scientific Presentations section of Concert’s website.

Gli interventi di riqualificazione hanno visto Andermatt arricchirsi di una nuova piazza cittadina, dominata dal nuovo auditorium, sulla quale si affacciano anche nuovi hotel, edifici residenziali e chalet.

Concert Pharmaceuticals Announces Advancement of Novel Drug Candidate in Schizophrenia

Posted by fidest press agency su domenica, 4 marzo 2018

Concert Pharmaceuticals, Inc. (NASDAQ: CNCE) today announced the next development candidate in its product pipeline with the selection of CTP-692, a novel drug for adjunctive treatment of schizophrenia, a devastating, chronic illness with significant unmet need. CTP-692 is a deuterated form of D-serine, an endogenous, required co-agonist of the N-methyl-D-aspartate (NMDA) receptor. NMDA receptor hypofunction is believed to contribute to the pathophysiology of schizophrenia and enhancement of D-serine levels is believed to benefit individuals with schizophrenia. CTP-692 is being developed as a potential adjunctive therapy to antipsychotic medicines with the potential to improve positive and negative symptoms as well as cognitive function in these patients. The Company intends to complete preclinical evaluation and advance CTP-692 into clinical development by year-end 2018.“We see neuropsychiatry as an area in which our team is well-positioned to broaden and expand our pipeline and apply our deuterium chemistry technology to create innovative medicines with differentiated therapeutic properties,” said Roger Tung, Ph.D., President and CEO of Concert Pharmaceuticals. “Our newest development candidate, CTP-692, represents an exciting opportunity to improve upon the existing standard-of-care with the potential to make a meaningful difference for patients with schizophrenia.”Despite its therapeutic potential as an adjunctive antipsychotic medication, the development of D-serine has been limited by safety concerns. D-serine has been shown to cause nephrotoxicity in preclinical testing. In addition, laboratory findings suggesting a possible renal safety signal were observed in some patients who received D-serine in clinical studies.CTP-692 has the potential to improve the safety profile of D-serine. In preclinical evaluation, Concert found that selective deuterium modification increased D-serine exposure and substantially reduced evidence of renal impairment. As a result, the Company believes that it can explore a wider exposure range to achieve optimal therapeutic levels in the clinic with a much lower risk of renal toxicity. These results support the further advancement of CTP-692.

Jewish and Arabic musicians in concert

Posted by fidest press agency su mercoledì, 8 luglio 2009

Eliya“When people of various Middle Eastern cultures come together around music, connection and love are fostered,” said Eliyahu Sills, the group’s leader. “I have been in rooms where Lebanese, Israeli, Egyptian, and Iranian people come together through melody and rhythm, all singing the same songs in different languages. This experience creates so much joy and connection that it can serve as a powerful tool for melting the ice of political animosity. We experience this joy and connection often at our concerts, and hope to share it with as many people as we can.” Rachel Valfer, the group’s lead vocalist adds, “its hard to hold a grudge against someone who loves something you love, who sits next to you clapping to the same rhythms and moved at the same shift in the maqam mode. We have entered a tense room and felt hostility melt around this musical experience.” “Eastern Wind” masterfully mixes the sounds of Middle Eastern instruments to create a collection of traditional and original songs that celebrate the shared musical and spiritual heritage of a region now deeply divided by ethnic and religious differences. Featuring folk tunes, improvisations, and songs of love and devotion from the 12th century onward, the eclectic ensemble uses traditional instruments to travel on a musical journey through Arabia, Armenia, Egypt, Israel, Morocco, Turkey and Yemen.
Qadim is a word found in both Arabic and Hebrew meaning ‘ancient’ as well as “that which will come.”  The Qadim ensemble plays music inspired by many traditions of the Near East, in an effort to bring the richness and beauty of these ancient sounds to contemporary audiences. Recognized for their spellbinding performances, Qadim (pronounced ka-deem) is comprised of an ensemble of acclaimed Bay Area musicians. The San Francisco Bay Area-based group has attracted a growing global audience who are responding to their authentic musicianship and timely message. Durme Durme, a track from the group’s debut CD, Songs of Qadim, was recently featured on Mediterraneo by Difference Music, an internationally distributed 4-CD set of 73 traditional and modern Mediterranean songs and 200-page hard-bound book showcasing the region’s history, geography and culture. The track Ya Adili Bilah can be heard on Petrol Records new collector’s edition compilation Arabia: The Greatest Songs Ever. Eliyahu Sills has been studying and performing music for over 20 years on many instruments including upright bass, bansuri (the bamboo flute of India), and the ney (the reed flute of the Middle East.) In the early 1990s, he studied the upright bass at The New School for Jazz and Contemporary Music in New York City. Deeply influenced by teachers  Makanda Ken McIntyre, Arnie Lawrence and Reggie Workman (John Coltrane, Art Blakey), Eliyahu began performing as a  sideman and bandleader in renowned jazz clubs such as The Village Gate and Small’s. In the years that followed,  Eliyahu fell in love with the flute and applied the musical theory that he learned through his jazz studies to teach  himself this most ancient of instruments. After falling under the spell of the Sufi music of Turkey, he became a  devoted student of both the ney and the bansuri. He has since continued to study and perform these ancient instruments under the guidance of his teacher, bansuri master  G.S. Sachdev, to whom he is a devoted student. (Eliya)
